Roma’s Gianluigi Buffon sealed the win with a second.
Juventus have won seven straight games and have a chance of making it six in a row with a win over Roma.
This was their fourth victory in six UEFA Europa league matches.
They are second in the group after drawing 1-1 with Barcelona in their opening group game on Sunday.
Juve had the best chances in the first half and were denied by a poor save by Roma goalkeeper Gianluisi Buffon.
In the second half, Juventus had the better chances and Mandzakic struck a second to make things 2-0.
They have scored four goals in their past five Champions League matches.
Rome have won three of their past four games against Juventus.
